What Is Retatrutide?
Retatrutide (LY-3437943) is an investigational triple receptor agonist peptide that activates three metabolic receptors at once: GLP-1 (glucagon-like peptide 1), GIP (glucose-dependent insulinotropic polypeptide), and glucagon. The combined action of all three pathways is the basis for the compound being studied as one of the most potent investigational research compounds in this class.
In Phase 2 clinical research published in The New England Journal of Medicine in 2023, retatrutide reported approximately 24.2% body weight reduction at 48 weeks at the highest study dose. Published comparator research reports approximately 22.5% body weight reduction for tirzepatide (dual receptor agonist) and approximately 14.9% for semaglutide (single receptor agonist) in their respective trials. Phase 3 trials under the TRIUMPH program are due to read out from late 2026.

How Retatrutide Works — Triple Receptor Mechanism
